SNARF-1, AM is a cell-permeant pH probe with a pKa of 7.5 after hydrolysis of the acetoxymethyl ester of the probe upon entering live cells. SNARF-1 is a ratiometric indicator sensitive to measuring pH changes between pH 7 and pH 8 and shifts from 580 nm under acidic pH to 640 nm under basic conditions. It also undergoes a shift in its excitation peak between pH6 to pH9 from ~532 nm at pH6 to ~561nm at pH9, but typically this probe is typically used in fluorescent imaging applications with a single excitation peak at 555nm to monitor the dual emission shift.
