	PKC theta	Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases that can be activated by calcium and the second messenger diacylglycerol. PKC family members phosphorylate a wide variety of protein targets and are known to be involved in diverse cellular signaling pathways. PKC family members also serve as major receptors for phorbol esters, a class of tumor promoters. Each member of the PKC family has a specific expression profile and is believed to play a distinct role. The protein encoded by this gene is one of the PKC family members. It is a calcium-independent and phospholipid-dependent protein kinase. This kinase is important for T-cell activation. It is required for the activation of the transcription factors NF-kappaB and AP-1, and may link the T cell receptor (TCR) signaling complex to the activation of the transcription factors. 		Detection of Human PKC theta by Simple Western^TM. Simple Western lane view shows lysates of Jurkat human acute T cell leukemia cell line, loaded at 0.2 mg/mL. A specific band was detected for PKC theta at approximately 82 kDa (as indicated) using 5 µg/mL of Mouse Anti-Human PKC theta Monoclonal Antibody (Catalog # MAB4368). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.
		Detection of Human PKC theta by Western Blot. Western blot shows lysates of Jurkat human acute T cell leukemia cell line. PVDF membrane was probed with 2 µg/mL of Human PKC theta Monoclonal Antibody (Catalog # MAB4368) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for PKC theta at approximately 82 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 3.
		Detection of PKC theta in Jurkat Human Cell Line by Flow Cytometry. Jurkat human acute T cell leukemia cell line was stained with Human PKC theta Monoclonal Antibody (Catalog # MAB4368, filled histogram) or isotype control antibody (Catalog # MAB003, open histogram), followed by Phycoerythrin-conjugated Anti-Mouse IgG F(ab')₂ Secondary Antibody (Catalog # F0102B). To facilitate intracellular staining, cells were fixed with paraformaldehyde and permeabilized with methanol.
