About p53 and CoraFluor-1
p53 This gene encodes a tumor suppressor protein containing transcriptional activation, DNA binding, and oligomerization domains. The encoded protein responds to diverse cellular stresses to regulate expression of target genes, thereby inducing cell cycle arrest, apoptosis, senescence, DNA repair, or changes in metabolism. Mutations in this gene are associated with a variety of human cancers, including hereditary cancers such as Li-Fraumeni syndrome. Alternative splicing of this gene and the use of alternate promoters result in multiple transcript variants and isoforms. Additional isoforms have also been shown to result from the use of alternate translation initiation codons from identical transcript variants (PMIDs: 12032546, 20937277). CoraFluor-1 CoraFluors are luminescent terbium complexes developed by the Mazitschek lab at the Massachusetts General Hospital and Harvard University. The high brightness, long emission lifetimes (> 1 ms) and excellent stability of CoraFluors make them well-suited tools for biomedical assay applications, including homogenous and intracellular time-gated fluorescent measurements (e.g., homogenous and intracellular TR-FRET assays). CoraFluors can be excited with near-UV light and exhibit several narrow emission bands (490 nm, 545 nm, 585 nm, 620 nm), providing compatibility with a broad range of organic fluorophores and fluorescent proteins in TR-FRET experiments.
