About Perforin and PE-Cy7
Perforin This gene encodes a protein with structural similarities to complement component C9 that is important in immunity. This protein forms membrane pores that allow the release of granzymes and subsequent cytolysis of target cells. Whether pore formation occurs in the plasma membrane of target cells or in an endosomal membrane inside target cells is subject to debate. Mutations in this gene are associated with a variety of human disease including diabetes, multiple sclerosis, lymphomas, autoimmune lymphoproliferative syndrome (ALPS), aplastic anemia, and familial hemophagocytic lymphohistiocytosis type 2 (FHL2), a rare and lethal autosomal recessive disorder of early childhood. PE-Cy7 PE-Cyanine®7 (PE-Cy7, RPE-Cy7) is a far red-emitting tandem fluorophore that combines phycoerythrin (PE) and Cy7. The donor molecule, PE can be excited by the 488-nm blue, 532-nm green, or 561-nm yellow-green laser and and transfers energy to the acceptor molecule, Cy7, which emits light that can be captured with a 780/60 nm bandpass filter. PE-CY7 has an excitation peak at 565 nm and an emission peak at 778 nm, and is a suitable alternative to PE-Vio®770 (Miltenyi) and cFluor BYG781 (Cytek).
