The OKT8 monoclonal antibody specifically recognizes CD8 alpha (CD8a or CD8α). CD8a is a ~32-34 kDa type I transmembrane glycoprotein that is encoded by CD8A, which belongs to the immunoglobulin superfamily (IgSF), and is commonly referred to as CD8. CD8a is expressed by the majority of thymocytes, subpopulations of αβ T cells and γδ T cells, and by subsets of NK cells. Cell surface CD8a is expressed either as a disulfide-linked homodimer (CD8αα) or as a heterodimer (CD8αβ) when disulfide-bonded to a CD8 beta chain (CD8b or CD8β). CD8-positive αβ T cells coexpress both CD8αα homodimers and CD8αβ heterodimers whereas some γδ T cells and NK cells express CD8αα homodimers. CD8 plays important roles in T cell activation and selection. The extracellular IgSF domain of CD8α binds to a non-polymorphic determinant on HLA class I molecules (α3 domain) and enables CD8 to function as a coreceptor with MHC class I-restricted TCR during T cell recognition of antigen. The cytoplasmic domain of CD8α associates with Lck, a Src family protein tyrosine kinase that is involved in intracellular signaling. The OKT8 antibody reportedly crossreacts with CD8a expressed by leucocytes from some non-human primate (NHP) species. |

The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system. The CD8 antigen acts as a coreceptor with the T-cell receptor on the T lymphocyte to recognize antigens displayed by an antigen presenting cell in the context of class I MHC molecules. The coreceptor functions as either a homodimer composed of two alpha chains or as a heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains. This gene encodes the CD8 alpha chain. Multiple transcript variants encoding different isoforms have been found for this gene. The major protein isoforms of this gene differ by the presence or absence of a transmembrane domain and thus differ in being a membrane-anchored or secreted protein. [provided by RefSeq, May 2020]
Brilliant™ Violet 711 (BV711) is a red-emitting tandem fluorophore that can be excited by the 405 nm Violet laser and collected using a 710/50 bandpass filter. BV711 has an excitation peak at 405 nm and an emission peak at 711 nm. BV711 exhibits a medium level of brightness and is most often used in flow cytometry, specifically, it is an alternative to nanocrystals used for intracellular flow. This dye is part of the Brilliant™ Violet dye line of fluorescent polymers. Brilliant™ Violet 421 polymer is employed as the donor molecule in a series of tandem dyes with acceptor molecules emitting at various points across the visible light spectrum. The Brilliant™ Violet dyes are a superior alternative to QDot nanocrystals and similar to SuperNova dye from Beckman Coulter and StarBright dyes from Bio-Rad.
