Description Clone REAL487 is an antibody fragment derived from the full CD34 antibody molecule. It displays no binding to Fc receptors. The recombinantly engineered antibody fragments are multimerized to form the REAdye_lease Complex to bind markers with high avidity. | Clone REAL487 recognizes the human CD34 antigen, a single chain transmembrane glycoprotein, expressed on human hematopoietic stem and progenitor cells, endothelial progenitor cells, vascular endothelial cells, embryonic fibroblasts, and some cells in fetal and adult nervous tissue. The antigen is absent on fully differentiated hematopoietic cells such as normal peripheral blood lymphocytes, monocytes, granulocytes, erythrocytes, and platelets. CD34 antibodies can be used for studies of hematopoiesis and nonhematopoiectic stem cells, phenotyping of endothelial cells, and endothelial progenitor cells (EPCs), hematopoietic stem cells, and studies on phenotyping of hematologic malignancies, e.g. the identification of blasts in leukemia. Additionally it is a marker for solitary fibrous tumors and gastrointestinal stromal tumors. | For removal of REAdye_lease fluorochromes for optional relabeling with different fluorochrome-conjugated REAdye_lease antibodies use the REAlease Support Kit (130-120-675).
About CD34 and FITC
CD34 The protein encoded by this gene may play a role in the attachment of stem cells to the bone marrow extracellular matrix or to stromal cells. This single-pass membrane protein is highly glycosylated and phosphorylated by protein kinase C. Two transcript variants encoding different isoforms have been found for this gene. FITC Fluorescein isothiocyanate (FITC) has an excitation peak at 495 nm and an emission peak at 519 nm. The name FITC is a misnomer in that the isothiocyanate is a reactive form of this dye. Once FITC is conjugated to an antibody, it is simply Fluorescein conjugated. FITC is one of the most widely used dyes for fluorescent applications, therefore most instruments come standard with a 488 nm laser and FITC filter set up. FITC is commonly conjugated to secondary antibodies and used in applications such as flow cytometry, immunocytochemistry, and immunohistochemistry. FITC is relatively dim, sensitive to photobleaching and it is susceptible to changes is pH. There are better performing alternatives to FITC, like Vio®Bright 515, Alexa Fluor™ 488, iFluor® 488, CF®488A and DY-488. FITC is a long-time generic dye with no sole manufacturer or trademark.
