About HIF-1 beta and Alexa Fluor 555
HIF-1 beta This gene encodes a protein containing a basic helix-loop-helix domain and two characteristic PAS domains along with a PAC domain. The encoded protein binds to ligand-bound aryl hydrocarbon receptor and aids in the movement of this complex to the nucleus, where it promotes the expression of genes involved in xenobiotic metabolism. This protein is also a co-factor for transcriptional regulation by hypoxia-inducible factor 1. Chromosomal translocation of this locus with the ETV6 (ets variant 6) gene on chromosome 12 have been described in leukemias. Alternative splicing results in multiple transcript variants. Alexa Fluor 555 Alexa Fluor™ 555 (AF555, Alexa 555) has an excitation peak at 555 nm and an emission peak at 565 nm, and is spectrally similar to Cy®3 (GE Healthcare), TRITC, iFluor® 555 (ATT Bioquest) and iFluor® 560 (ATT Bioquest). Many flow cytometers are equipped with 532nm lasers, so while Alexa 555 can be used for flow cytometry, PE is more commonly used due to its brightness and spectral characteristics. This dye is commonly used for fluorescence microscopy applications.
