Description | Clone REA338 recognizes the human octamer-binding transcription factor 3/4 isoform A (Oct3/4A) and the mouse Oct3/4. Oct3/4 is a homeodomain transcription factor belonging to the POU transcription factor family and is also known as POU domain class 5 transcription factor 1 (POU5F1). Oct3/4 is critical for the maintenance of totipotency in primate blastomeres and for pluripotency of the inner cell mass of developing mammalian embryos. This critical role of Oct3/4 is maintained in embryonic stem cells, the developing germline, and germline tumors, such as seminomas and embryonic teratocarcinomas. When inducing pluripotency in differentiated cells, Oct3/4 appears to be an essential, but not the only required transcription factor. Oct3/4 acts as a transcriptional activator of genes involved in maintenance of undifferentiated state and as a repressor of differentiation-specific genes.In human it encodes two chief isoforms that are generated by alternative splicing – Oct3/4A and Oct3/4b. Only the Oct3/4A protein consequently contains the unique N-terminal transactivation domain that regulates DNA binding of Oct3/4A to target promoters. It is only this full length Oct3/4A protein – that localizes to the nucleus, binds to Oct3/4-regulated promoters, and contributes to the maintenance of pluripotency and self-renewal. | Additional information: Clone REA338 displays negligible binding to Fc receptors. |
