Brilliant Violet™ 421 (BV421) is a violet-emitting non-tandem polymer fluorophore that can be excited by the 405 nm violet laser and collected using a 450/50 bandpass filter. BV421 has an excitation peak at 405 nm and an emission peak at 421 nm, and is spectrally similar to Alexa Fluor 405 and Cascade Blue. Other dyes that are considered similar include V450 (BD Biosciences), VioBlue® (Miletnyi Biotec), Cascade® Blue (ThermoFisher Scientific), Pacific Blue (ThermoFisher Scientific), SuperBright 436 (ThermoFisher Scientific), and Alexa Fluor® 405 (ThermoFisher Scientific). BV421 is very bright and is most commonly used for flow cytometry. Its photostablity, fixation stablity, and non-toxicity properties also make BV421 useful for cell sorting and live cell imaging applications. This dye is part of the Brilliant Violet dye line, and this dye specifically is one of the most popular in the line. The Brilliant Violet dye line itself is very popular because other dyes in the line also use the Brilliant Violet 421 core as the donor molecule, while varied acceptor molecules emit light at various points across the visible light spectrum. This allows for multiple dyes to be compatible while excited by the single laser. In cytometry, a user may populate all the available channels off their 405-violet laser with Brilliant Violet dyes.
